  • How should precipitates in undiluted GelNest™ Basement Membrane Matrix be handled?

    Centrifuge at low speed at 4°C and remove the precipitate.

  • How should unused GelNest™ Basement Membrane Matrix be stored?

    GelNest™ Basement Membrane Matrix that has already been mixed with cell culture medium or buffer but remains unused should not be stored for reuse.

  • Can GelNest™ Basement Membrane Matrix be stored at -70°C?

    Yes. GelNest™ Basement Membrane Matrix can be stored at -70°C. It is recommended to aliquot the entire bottle into polypropylene tubes or other tubes made of materials suitable for ultra-low-temperature storage for convenient storage and use.

  • Does GelNest™ Basement Membrane Matrix exhibit autofluorescence?

    GelNest™ Basement Membrane Matrix is a protein mixture, and its protein components may contribute to fluorescence. When fluorescence is used to evaluate cell growth, establish an appropriate control and compare signals at the required wavelength to distinguish background fluorescence.

  • How should cells cultured in GelNest™ Basement Membrane Matrix be fixed for sectioning, immunohistochemistry, or immunofluorescence, and how can depolymerization be prevented?

    Fix the samples with 2% paraformaldehyde. To prevent depolymerization after fixation, 1% glutaraldehyde may be added. Glutaraldehyde is commonly used as a fixative for electron microscopy. However, when samples are intended for immunofluorescence, glutaraldehyde can produce substantial background fluorescence. To address this, quench the samples with NaBH₄ after fixation. NaBH₄ readily generates bubbles, so perform this step carefully on a level work surface, avoid agitation, and minimize bubble formation. Alternatively, use a lower concentration of glutaraldehyde, such as 0.1–0.5%; lower concentrations generally produce less background fluorescence.
